Simon de Vos was born in Antwerp on 20 October 1603, the son of the dice maker Herman de Vos and Elisabeth van Oppen. At age twelve, in 1615, he began five years of training with the portrait painter Cornelis de Vos, to whom he was not related, and in 1620, at seventeen, he was admitted as a master in the Antwerp Guild of Saint Luke. Some art historians have suggested he may afterward have spent time in Rubens's studio, or traveled abroad, possibly to Italy, but no archival record confirms either path, and the point remains conjecture rather than documented fact.

De Vos worked his entire career in Antwerp. His early output favored small, Caravaggesque genre scenes of merry companies of drinkers and smokers, of the kind now held by the Louvre. He later turned to religious and historical subjects painted close to Rubens's own style, several of which hung in Antwerp's churches, including the Cathedral, the Church of St Andrew, and the Abbey of St Michael, and were, according to an older secondary account of uncertain reliability, at times mistaken for Rubens's own paintings. He also collaborated with the flower painter Daniel Seghers, supplying the figures for garland still lifes. Simon de Vos died in Antwerp on 15 October 1676, at the age of seventy-two.
